My first hard drive is a 1TB WD CAVIAR BLACK WD1002FAEX, SATA 6 Gb/s, 64MB CACHE (7200rpm)
would it be advantageous to have a partition say of 250MB for the operating system?

250 "MB"? You'd need at least a 20GB partition in my opinion for Windows 7... However, there won't be any advantages to partioning your hard-drive other than being more organised (which isn't a bad thing).

There are no advantages of partitioning, unless it's beneficial for you personally to have two virtual hard drives for storage.

Defrag is largely redundant is using the NTFS file system, there is no logical reason for the OS speed to be affected as its only a partition.
