First time buyer on here - couple of questions on what to expect...

When it arrives will the operating system already be installed? I ordered a SSD (240GB) and HDD (1TB), will the operating system be installed on the SSD automatically or should I reach out to PCS to confirm? Is there any point in partitioning either of these drives? What will this be like out of the box? Is there much set up involved? Does it take long to get it all up and running? I don't mean actually hooking it up but rather when I turn it on for the first time. What can I expect day one basically?

The operating system and drivers will be installed.
All you should need to do is activate windows so be sure to take a note of the product key which will be on a sticker on the back or side of the computer case.
PCS normally install the operating system on "1st Hard Disk" you selected on the configurator.
EDIT: There may be some packaging to remove from inside the case,but as DED says they will make this clear,and if you ordered a wireless card the antennas will be in the welcome pack.

As vanthus says, it is as "ready to use" out of the box as much as possible, but this can vary depending on certain components. Anything that does need doing should have directions though.

(V! How could you! I'm pretty sure changing your avatar is one of the seven seals of the Apocalypse!)

The SSD will be your primary Hard Drive so your operating system will be preloaded onto this. Just remember that as this is your primary drive make sure to select what drive to save stuff too before you download SSD will be C Drive your Sata HDD will be an I Drive.
